Optimization and Pilot Test on the Fermentation Condition of D-Ribose

摘要 [目的]研究利用枯草芽孢杆菌的莽草酸营养缺陷型突变株发酵生产D-核糖的发酵条件。[方法]以枯草芽孢杆菌CB.sems-10为出发菌株,通过正交试验得出最佳培养基配方并利用种子罐和发酵罐对发酵条件进行了中试试验。[结果]单因素试验表明,接种量为10%时,D-核糖产率最高;发酵前期的适宜pH值约为7.2,发酵中后期的适宜pH值在偏酸性范围内;37℃下D-核糖的产率最高;摇床转速为170 r/m in时,D-核糖的产率最高。极差分析表明,各因素对D-核糖产率的影响依次为:酵母粉>(NH4)2SO4>玉米糖>Mn-SO4>玉米浆。最佳培养基配方为:玉米糖20%+酵母粉0.6%+(NH4)2SO40.5%+MnSO40.05%,最佳pH值为7.2。枯草芽孢杆菌CB.sems-10的适应能力强,转糖率高,其平均D-核糖产率为108.6 mg/L,最高达127.1 mg/L。[结论]该研究为利用枯草芽孢杆菌CB.sems-10发酵生产D-核糖奠定了理论基础。 [ Objective] The purpose was to study the fermentation condition of producing D-ribose through fermentation with shikimie acid auxotrophy mutant of Bacillus subtilis. [ Method] With B. subtilis CB. seres - 10 as original strain, the optimum medium formula was obtained through orthogonal experiment and the pilot test was performed on fermentation condition with seed tanks and fermentation tanks. [ Result] It was shown in the single factor experiments that when the inoeulatiov amount was 10% , the yield rate of D-ribose was highest; the suitable pH value was about 7.2 in earlier fermentation and it was in the range of weak acidity in middle and late fermentation ; the yield rate of D-ribose was highest at 37 ℃ and it was highest when the rotation speed was 170 r/min. The range analysis showed that the influences of factors on the yield rate of D-ribose were as follows: yeast powder 〉 (NH4)2SO4 〉 corn sugar. 〉 MnSO4 〉 corn syrup. The optinmm medium fornmla was corn sugar 20% + yeast powder 0.6% + (NH4)2SO4 0.5% + MnSO4 0.05% and the optimum pH value was 7.2. B. subtilis CB. sems- 10 bad strong adaptability and high glucose conversion rate and its average yield rate of D-ribose was 108.6 mg/L and its highest yield rate was up to 127.1 mg/L. [ Conclusion] The study laid a theoretical foundation for producing D-ribose through fermentation with B. subtilis CB. seres-10.
