大黄素诱导细胞凋亡对不同类型急性胰腺炎腺泡死亡方式的影响 被引量:4

Effect of emodin on pancreas aeinar death patterns in defferent types of acute pancreatitis

摘要 目的探讨大黄素(emodin)诱导细胞凋亡对不同类型急性胰腺炎(AP)腺泡细胞死亡方式影响及其可能机制。方法制作大鼠急性水肿型胰腺炎(AEP)与急性坏死型胰腺炎(ANP)模型,使用大黄素进行治疗。采用原位TUNEL及RTPCR等技术,观察胰腺组织内细胞凋亡、NF-κB mRNA、Bax mRNA及TNF-α mRNA等变化情况。结果各组大鼠胰腺细胞凋亡指数与胰腺组织病理学评分呈负相关;大黄素治疗可诱导各型AP模型腺泡细胞凋亡,胰腺病理学改变及组织学评分改善,胰腺组织内NF-κB mRNA及Baxm RNA表达增加。结论诱导不同类型AP胰腺细胞凋亡叮明显减轻AP病情。大黄素为治疗AP的有效药物,可能通过影响NF-κB活化及Bax的表达米诱导AP时腺泡细胞的凋亡,减轻胰腺炎的严重程度。 Objeetive To investigate the effect of Chinese medicine emodin on death patterns of panereaticacinar cells in different types of aute pancreatitis and explore its possible mechanism. Methods Rat acute edematous panerealitis models and acute necrotic pancreatitis models were estab lished and treated with emodin. Alternations of apoptosis, NF-κB mRNA, Bax mRNA and TNF-α mRNA expression in pancreatic tissue were studied with in situ TUNEL and RT- PCR. Results Negative correlation was found between apoptosis and the severity of pancreatic damage. Following the treatment with emodin, more apoptosis than necrosis was observed in all treaded groups of acute pancreatitis and the severity of pancreatie histological pathologic damage was dramatically improved, l.ocal expression of NF -κB mRNA and Bax mRNA increased. Conclnsion Pancreas acinar death patterns of apoptosis are induced by emodin. Emodin might regulate the acinar cell death patterns by changing the activation of NF-κB and the expression of bax to lessen the severity.
