摘要
目的研究亚低温对局灶性脑缺血再灌注损伤(fCIRI)后成年大鼠脑内内源性神经干细胞(eNSCs)及其分化的影响。方法采用线栓法模拟局灶性大脑中动脉阻塞制造成年大鼠fCIRI,持续脑缺血2 h后再灌注。将动物随机分为fCIRI常温组f、CIRI亚低温组。fCIRI亚低温组于再灌注时即开始实施亚低温,持续1 h。在fCIRI后1 d取脑,应用免疫荧光观察eNSCs和新生神经细胞的表达。结果与fCIRI常温组比较fCIRI亚低温组侧脑室下区中eNSCs数量有所增加(P<0.05),新生神经细胞与新生神经胶质细胞的比率较fCIRI常温组增多(P<0.05)。结论fCIRI后亚低温能促进eNSCs增殖并更多地向神经细胞分化。
Objective It is to study the influence of sub-hypothermia on endogenous neural stem cells (eNSCs) in adult rats after focal cerebral ischemic and reperfusion injury (fCIRI) and its' differentiation. Methods The fCIRI of adult rats were established with line fastening method modeling focal middle cerebral artery occlusion (MCAO). Reperfusion was done after cerebral ischemia abidingly for 2 h. The animals were randomly divided into fCIRI common temperature group and fCIRI sub-hypothermia group. The fCIRI sub-hypothermia group was given sub-hypothermia at the time of reperfusion for 1 h. The brain was taken after fCIRI for 1 d. The eNSCs and the expression of neoformative nerve cell were observed with immunofluorescence. Results Compared with fCIRI common temperature group, the quantity of eNSCs in SVZ in fCIRI subhypothermia group was increased somewhat (P 〈 0.05), the ratio between neoformative nerve cell and neoformative glial cell in fCIRI sub-hypothermia group was increased (P 〈 0.05). Conclusion Sub-hypothermia after fCIRI can stimulate the proliferation of eNSCs and more differentiation to nerve cell.
