摘要
目的:考察不同浓度甲醇对决明子中5种蒽醌类成分的含量的影响。方法:采用高效液相色谱法测定大黄中芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚含量,KromasilC18分析柱(250㎜×4.0㎜,5μm);柱温:室温;流动相:甲醇-0.05%磷酸水(75:25);检测波长:254nm;流速:1.0ml·min-1。结果:芦荟大黄素含量为0.2142mg·g-1~0.5725mg·g-1,大黄酸含量为0.0238mg·g-1~0.2044mg·g-1、大黄素含量为1.0122mg·g-1~2.9344mg·g-1、大黄酚含量为9.3577mg·g-1~24.4438mg·g-1、大黄素甲醚含量为0.3699mg·g-1~0.9551mg·g-1,平均回收率范围为98.10%~105.39%,平均RSD小于3%(n=6)。结论:不同浓度甲醇提取的大黄药材中5种蒽醌类化合物含量差异大,其中以80%甲醇甲醇提取效果最佳,纯甲醇提取效果最差。
Aim: Explore the effect of methanol of different concentration on the content of 5 anthraquinone compounds in cassiac torae. Method: adopt high efficiency liquid chromatography to determine the content of aloe-emodin, rhein, emodin, chrysophanol and physcione in rhubarb; Kromasil C18 analytical column; column temperature: room temperature; moving phase: methanol-0.05% H3PO4 (75:25) ; detection wave length: 254nm; flow rate: 1.0ml·min^-1.Result:aloe-emodin content is 0.2142mg·g^-1-0.5725mg·g^-1, rhein content is 0.0238mg·g^-1-0.20 44mg·g^-1, emodin content 1.0122mg·g^-1-2.9344mg·g^-1, chrysophanol content is 9.3577mg·g^-1-24.4438mg·g^-1, physcione content is 0.3699 mg·g^-1-0.9551mg·g^-1, the average recovery is 98.10%-105.39%, the average RSD is less than 3% (n=6) . Result: the content of 5 anthraquinone compounds in rhubard differs greatly which is extracted with mathnol with different concentration. Methnol with concentration of 80% has the best extraction effect while the pure methnol has the poorest extraction effect.
