摘要 [目的]研究猪血超氧化物歧化酶(SOD)与其他生物活性物质的联合提取工艺,为充分利用猪血资源和规模化生产SOD提供技术基础。[方法]从猪血中分离纤维连接蛋白、免疫球蛋白和血红素后,提取SOD。通过优化邻苯三酚的用量、SOD样液用量、反应温度和缓冲液pH值,对邻苯三酚自氧化法测定SOD活性的条件进行了优化。按照优化后的条件测定联合提取的SOD的比活力。[结果]SOD活性测定的优化条件为50 mmol/L邻苯三酚7μl,50 mmol/LTris-HCl(pH8.2)3 ml,反应温度为25℃,SOD样液添加量为10μl。联合提取的SOD的比活力达到5 056 U/mg蛋白质。[结论]通过现代生物工程集成技术从猪血中分级提取了4种生物活性物质,所提取的SOD具有较好的活性。 [ Objective ] To study the combined extracting process of porcine blood superoxide dismntase (SOD) and other bioaetive substances and thus to provide technical basis for making full use of blood resources and large-scale production of SOD. [ Method ] Fibronectin, immunoglobulin, and hemoglobin were isolated from porcine blood, and SOD was extracted. Trace pyrogalM self-oxidation method to determine SOD activity was modified by optimizing the volume of pyrogallol and SOD samples, reaction temperature, and buffer pH. The specific activity of SOD was determined with the optimized extraction conditions. [ Result] The improved experimental conditions of SOD activity detection were as follows: 7 μl pyrogallol (50 mmol/L), 3 ml Tris-HCl (50 mmol/L, pH8.2), reactive temperature at 25 ℃, and 10 μl SOD sample solution. The specific activity of extracted SOD was 5 056 U/mg protein. [ Conclusion] Four kinds of bioactive substance can be isolated from porcine blood by modem biological engineering integration technology, and the extracted SOD has better activity.
