茎瘤芥超氧化物歧化酶活性的研究 被引量:1

摘要 [目的]揭示茎瘤芥超氧化物歧化酶(SOD)的活性以及pH、底物浓度、温度对SOD活性的影响。[方法]以茎瘤芥的瘤茎及叶片为试验材料,用邻苯三酚自氧化法分别测得它们中SOD的活性,以及pH、底物浓度、温度对其活性的影响,从而比较茎瘤芥瘤茎及叶片中SOD的含量,以及是否具有开发利用的价值。[结果]茎瘤芥瘤茎SOD的活性在200~300 U/g;茎瘤芥叶片SOD的活性在250~400U/g。说明茎瘤芥叶片SOD活性高于茎瘤芥瘤茎SOD活性。茎瘤芥SOD在pH为8.3~8.6有最大酶活性;最适温度为50℃;最适底物浓度为7 mmol/L左右。[结论]茎瘤芥叶片SOD活性高于茎瘤芥瘤茎SOD活性。研究结果为开发茎瘤芥资源提供了参考。 [Objective] The activity of SOD of Brassica juncea Cross.var.tumida Tsen et Lee and the effect of the pH value,the concentration of substrate and temperature on it were explored.[Method] The activity of SOD,the effect of the pH value,concentration of substrate and temperature on it of the tumorous stem and leaf of Brassica juncea Cross.var.tumida Tsen et Lee as experimental material were measured with the method of pyrogallicacid oxidize and the value of its developing and using was analyzed.[Result] The activity of SOD in the tumorous stem of Brassica juncea Coss.var.tumida Tsen et Lee was between 200 U/g and 300 U/g and in the leaf,between 250 U/g and 400 U/g,which indicated that the activity of SOD in the leaf was higher than that in the tumorous stem.The greatest activity of SOD in the plant was under the condition of pH between 8.3 to 8.6;under the optimum temperature of 50 ℃ and the most suitable concentration of substrate of around 7 mmol/L.[Conclusion] The activity of SOD in the leaf of tumour stem mustard was higher than that in the tumorous stem and the result could provide the reference for the development utilization of tumour stem mustard.
