摘要
从广西五味子中分离荧光物质并建立用荧光法测定南五味子中安五脂素含量的方法。以乙醇提取,乙醚萃取并用硅胶柱色谱分离纯化,采用理化性质和波谱分析鉴定化合物的结构;研究它们的荧光性质,用荧光法和高效液相色谱法同时测定南北五味子中安五脂素的含量。从广西五味子分离纯化出4个化合物,分别为安五脂素、五味子甲素、五味子酯甲和原儿茶酸;安五脂素的荧光性最强,五味子酯甲没有荧光性;南五味子乙醇提取液的荧光光谱与安五脂素一致;南五味子中安五脂素用荧光法和高效液相色谱法分析结果无明显的差异,而北五味子荧光法测定结果偏高。荧光法具有灵敏度高、选择性好等特点,可直接用于南五味子中安五脂素的定量分析。
Objective Fluorescence compounds are isolated in Schisandra chinensis from Guangxi and a method is established for the determination of anwulignan in Kadsura by fluorospectrophotometry. Compounds are isolated from ethanolie extract by silica gel column chromatography and their structures were identified by physicochemical properties and spectroscopic analysis. The fluorescence properties of compounds are investigated. The content of anwulignan is determined by HPLC and fluorospec-trophotometry in Schisandra shinensis 4 compounds are obtained and identified as anwulignan (I), deoxyschisandrin (II), sehisantherin A(III) and protocatechuie acid(IV), respectively. The fluorescence property of anwulignan is the strongest and no fluorescence is produced by schisantherin A under the same condition. Fluorescence spectra are almost same between ethanolic extract of Kadsura and anwulignan. The content of anwulignan in Kadsura by fluorescence spectrophotometry isn't obvious difference with that by HPLC in precision and accuracy. However, the content in Schisandra by fluorescence spectrophotometry is higher than that by HPLC. The fluorescence method is sensitive and good selectivity. It is successfully applied to quantitative analysis of anwulignan in Kadsura.
