摘要
全合成甘露糖化壳聚糖,为靶向递送药物至巨噬细胞和树突状细胞提供载体材料。实验以甘露糖为起始原料,通过6步反应合成预期取代度为5%~10%的甘露糖化壳聚糖,分别用红外(FTIR)、元素分析等技术对其结构进行确证和表征,并通过HeLa和A549细胞毒性试验评价其细胞毒性。结果表明,所制备的甘露糖化壳聚糖为橘黄色固体,不溶于水,在pH低于6.0的醋酸盐缓冲液中溶解良好,取代度为7.35%;其细胞毒性较低,当浓度达到100μg/ml时,细胞存活率大于80%。合成的甘露糖化壳聚糖是一种安全的靶向递送载体。
In order to study the nanoparticle delivery system targeting macrophage and dendritic cells, Mannosylated chitosan (MC) with the expected degree of substitution to be 5%-10% was synthesized and characterized via FTIR and elemental ana!ysis. Cytotoxicity was evaluated in vitro via Hela cells and A549 cells MTT tests. It is found that the product is deep yellow solid with the degree of substitution to be 7.35%, insoluble in water while soluble well in acetic acid buffer (pH〈6.0), and the cyto- toxicity is low due to the fact that cell viability is higher than 80 % when the concentration of MC inereases to 100μg/ml. MC is successfully synthesized and safe to delivery of biological molecules.
