摘要
对传统转瓶进行了改进,使之成为一套既可有效控制供氧又可在线检测细胞摄氧率的小型多功能培养装置,并在此基础上,系统考察了不同供氧状况对IPLB-Sf21-AE昆虫细胞生长和代谢的影响。结果表明:维持适宜的溶氧水平对优化昆虫细胞的生长和代谢至关重要,昆虫细胞的摄氧率与其生长代谢密切相关,并可作为在线监控细胞生理状态的重要依据。
The oxygen supply in spinner flasks is a principal limiting factor in highdensity cultivation of insect cells. An improvement was made on a conventional spinner flask, changing the flask into a benchscale multifunctional culture vessel in which the dissolved oxygen level could be effectively controlled and the oxygen uptake rate be monitored on line. The effect of different oxygen supply conditions on the growth of IPLBSf21AE (Sf21) insect cells were investigated by using the modified device. It was shown that the appropriate dissolved oxygen level was extremely important for optimizing the growth of insect cells. Moreover, the OUR of Sf21 was found to be strongly related to cell growth. Consequently, OUR could be regarded as an important parameter for monitoring and control of the physiological state of cell cultures.
