对几种相关酶联合固定化初探 被引量:2

Studies on Coimmobilization of α-amylase, Glucoamylase and Glucose Isomerase

摘要 用聚丙烯酰胺包埋,再以戊二醛交联联合固定了α淀粉酶、糖化酶和葡萄糖异构酶,对联合固定化的最适条件和联合固定化酶的性质进行了初步研究.结果表明联合固定化酶较固定化单酶更能发挥协同效应,能够将底物一步水解并转化为产物.文中所用的固定化方法与其他方法相比,固定化酶的稳定性、半衰期及酶固定化后活力均较高.在60℃、pH55时,α淀粉酶和糖化酶二酶的联合固定化酶的活力最高,其操作半衰期达到79h;在60℃、pH75时,α淀粉酶、糖化酶和葡萄糖异构酶三酶的联合固定化酶的活力最高,其操作半衰期达到90h. α -amylase,Glucoamylase and Glucose Isomerase are coimmobilizaed by using both polyacrylamid embeding and glutaraldehyde crosslinking.The Optimal conditions for coimmobilization are established and the properties of the coimmobilized enzymes are studied.It has been found that coimmobilized enzymes has better synergism compared with separately immobilized enzymes and that starch could be directly converted into glucose and even fructose by coimmobilied enzymes.It has also been found that the stability,half life and activity of the coimmobilized enzymes obtained by both polyarcylamid embeding and glutaraldehyded crosslinking are higher than those of enzymes made with diazo method.When the temperature is 60℃ and the pH value is 5.5,the coimmobolized α- amylase and glucoamylase show the highest activity,the half life of the coimmobilied enzymes is 79h,and at the same temperature with a pH Value 7.6,the coimmobilized amylase,Glucoamylase and Glucose Isomerase exhibit the highest activity and the half life of the coimmobilied enzymes is 90 h.
