摘要
目的建立反相高效液相色谱法(RP-HPLC)测定血清中的茶碱浓度的方法。方法以苯巴比妥为内标,血清样品用二氯甲烷提取,采用Venusil XBP-C18柱(4.6×200mm,5μm)分析。流动相为甲醇:水(50:50),流速1.0mL.min-1,检测波长275nm,柱温25℃。结果本法在6.25~25μg.mL-1间线性关系良好。血清中茶碱的标准曲线方程为y=1.1386x-0.8262,r=0.9991,RSD1.18%(n=5),最低检测质量浓度为0.31μg.mL-1。日内、日间精密度RSD均小于5%。茶碱的回收率为91.89%~105.15%。结论该方法灵敏度高、准确、重复性好,可以用于常规茶碱血药浓度的测定。
OBJECTIVE To establish a simple rapid RP-HPLC method for the determination of theophylline concentration in serum. METHODS With Phenobarbital as internal standard, the plasma sample was extracted with methylene dichloridprotein. The seperation was carried out using Venusil XBP-C18(4.6 × 200mm, 5μm) column with the mobile phase of methanol-H20 (50 : 50 ). The flow rate was 1.0mL·min^-1. The detection wave length was 275 nm. The column temperature was 25℃ . RESULTS The linear range was 6.25- 25μg·mL^-1. The correlation coefficient was 0. 9991 (RSD = 1.18 %, n = 5). The lowest detecting concentration was 0.31μg·mL^-1.The intra-day and inter-day precision were both less than 5 %. The recovery rate was between 91.89- 105.15 %. CONCLUSION The method which is sensitive, accurate, and has good reproducibility could be used as general method to determine Theophylline in plasma.
