Immunofluoresence histochemical double staining technique was used to qualitively examine the distributions and relationships between Bandeiraea Simplicifolia Isolectin B 4 (BSI B 4) labeled primary afferent C fibers and terminals and substance P (SP) , calcitonin gene related peptide (CGRP) , phosphate activated glutaminase (PAG, which is a specific marker for glutamatergic neurons) , nitric oxide synthase (NOS) like immunoreactive (LI) fibers and terminals in the superficial laminae of the trigeminal subnucleus caudalis (Vc) and dorsal horn of the spinal cord. SP/BSI B 4, CGRP/BSI B 4, PAG/BSI B 4, and NOS/BSI B 4 double labeled cells in the trigeminal ganglion (TG) and dorsal root ganglion (DRG) were also quantitatively examined. The mainly overlapping area of SP , CGRP , PAG , and NOS LI fibers and terminals and BSI B 4 labeled C fibers and terminals was in the outer part of laminae Ⅱ (Ⅱo). In the TG and DRG, some BSI B 4 labeled cells which give off C fibers also showed SP , CGRP , PAG , and NOS LI. Among those double labeled cells, the number of PAG/BSI B 4 double labeled cells was the most numerous. Present results indicate that most of the primary afferent C fibers and terminals contain glutamate (Glu) and Glu is the most important neural active substance for the primary afferent C fibers. Additionally, some primary afferent C fibers also contain SP, CGRP, and NO.
