摘要
以茎尖诱导形成的原球茎为外殖体,采用液体培养方式探讨了不同浓度的激素配比、蔗糖浓度和添加不同量的新鲜液汁对蝴蝶兰原球茎增殖的影响,并比较了液体和固体两种培养方式的原球茎增殖特征。结果表明:不同浓度的外源激素及其配比对蝴蝶兰原球茎增殖影响不明显,但对原球茎生长影响较大,6-BA2.0mg/L+Ad1.0mg/L+NAA0.5mg/L的激素浓度组合比较适合蝴蝶兰原球茎增殖,蔗糖浓度对蝴蝶兰原球茎增殖和生长影响较大,适合蝴蝶兰PLBs增殖的蔗糖浓度为20g/L,添加10%~40%的新鲜椰汁对蝴蝶兰原球茎增殖生长有明显促进作用;原球茎液体增殖培养的时间不宜超过4周,5周内连续培养增殖曲线呈倒“V”字形;液体增殖培养的原球茎分化成苗的时间相对较晚。
The phalaenpsis PLBs( protocorm-like bodies) induced from stem tips were used as explants to be cultured in the liquid media which contain hormones, sucrose and coconut pahn juice. The effects of different concentrations of hormones, sucrose and coconut palm juice on PLBs propagation were studied in this experiment. Under the same condition, the character of PLB propagation in the liquid culture and solid culture were studied too. The results showed that the concentrations and combinations of hormones have a few effects on phalaenpsis PLBs propagation, but have great effect on PLB growth in the liquid culture. The best combination of hormones was 2.0 mg/L 6-BA + 1.0 mg/L Ad + 0.5 mg/L NAA. The concentrations of sucrose also have great effect on PLBs propagation and growth. The suitable medium for PLBs propagation in the liquid cuhure was 20 g/L. The medium added 10 % -40 % (v/v) coconut palm juice can not only elevate PLBs propagation coefficient but also improve the quality of PLBs. The propagation time of PLBs in the liquid culture was best within 5 weeks. The best medium for phalaenpsis PLBs propagation was 1/2MS + 2.0 mg/L 6-BA + 1.0 mg/L Ad + 20 g/L sucrose + 10 % (v/v) coconut palm juice. The PLBs growth curve showed collapsed "V" shape in this liquid medium within 5 weeks. The time of shoots regenerated from PLBs cultured in the liquid medium was longer than PLBs cultured in the solid medium.
