一株虫生真菌的分离鉴定及杀虫活性初探 被引量:7

摘要 [目的]为虫生真菌在害虫防治中的应用提供参考。[方法]以从罹病昆虫体表分离的虫生真菌Ekks1为供试菌种,对其进行形态鉴定;以ITS4和ITS5为引物,对Ekks1的ITS序列进行PCR扩增、电泳检测和序列分析,并检测Ekks1对小菜蛾和豆蚜虫的杀虫效果。[结果]经形态鉴定,Ekks1为半知菌亚门,丝孢纲,丝孢目,丝孢科,白僵菌属,球孢白僵菌(Beauveria.bassiana);经PCR扩增,成功获得了Ekks1菌株的ITS1-5.8s-ITS5 rDNA序列,经分析,ITS1-5.8s-ITS5序列为555 bp,菌株Ekks1与GenBank中B.bassiana(EF672309)的同源性为99%;菌株Ekks1在孢子浓度为1.5×107个/ml时,感染8 d后小菜蛾的校正死亡率为76%,感染72 h后豆蚜虫的校正死亡率为61%。[结论]Ekks1为白僵菌属真菌,其孢子浓度为1.5×107个/ml时,对小菜蛾和豆蚜虫均具有一定的致死效果。 [Objective] The aim was to provide the reference for the application of entomogenous fungi in pest control.[Method] With the isolated entomogenous fungi Ekks1 from the surface of infectious sect as the tested strain,its morphology was identified and its ITS sequence was amplified with ITS4 and ITS5 as primers,and the amplified ITS sequence was detected by electrophoresis and was analyzed.The insecticidal effects of Ekks1 on Plutella xylostella and Cowpea aphid were determined.[Result] Through morphological identification,Ekks1 was Beauveria.bassiana,Beauveria Bassiana,Hyphom ycetaceae,Hyphom ycetales,Hyphom ycetales,Feuterom ycotina.Through PCR amplification,the ITS1-5.8s-ITS5 rDNA sequence of Ekks1 strain was successfully obtained.After analysis,the length of ITS1-5.8s-ITS5 sequence was 555 bp,the homology of Ekks1 strain and B.bassiana(EF672309) in GenBank was 99%.When the concn.of Ekks1 was 1.5×107 spores/ml,the corrected mortality of P.xylostella was 76% after 8 d of infection and the corrected mortality of C.aphid was 61% after 72 h of infection.[Conclusion] Ekks1 was Beauveria fungi,and it had certain lethal effect on P.xylostella and C.aphid at 1.5×107 spore/ml.
