摘要
通过根尖染色体鉴定、田间农艺性状观测、桔梗皂苷-D含量测定和比较、蛋白质含量测定和蛋白质聚丙烯酰胺凝胶电泳实验,对桔梗同源四倍体进行了遗传稳定性研究。结果表明:桔梗四倍体株系的染色体数目仍为36条,农艺性状保持了典型的多倍体特征,桔梗皂苷-D含量及蛋白质含量均高于对照二倍体株系,各株系蛋白质电泳谱带基本相似。实验结果说明桔梗同源四倍体具有较好的遗传稳定性,为药用植物多倍体育种技术的发展和生产应用提供了依据。
Genetic stability on autotetraploid plant chromosome identification, agronomic characters of Platycodon grandiflorum was studied by means of observation, Platycodin D content determination and comparison, protein content determination and pro-PAGE. The results indicated that chromosome number in autotetraploid lines remained 36, agronomic characters showed typical autotetraploid characteristics, the content of Platycodin 13 and protein in autotetraploid lines were higher than that of the control diploid line and the pro-PAGE demonstrated the similarity among all the lines. The overall studies illustrated that autotetraploid plant of Platycodon grandiflorum had sound genetic stability,which can then provide a foundation for the development and application of Polyploid breeding technique of medicinal plant.
