摘要
以兴37和中育2号2个西瓜品种的子叶为外植体,对离体培养和植株再生进行了研究。结果表明,4个不同激素配比中附加5.0mg·L-16-BA+0.5mg·L-1IAA的培养基诱导芽率高达89.7%多,转接到KT0.2mg·L-1的MS培养基上进行再生芽的伸长培养后,在MS+0.1mg·L-1NAA的根诱导培养基中的诱导率达88.2%,其高于在附加0.5mg·L-1的IAA培养基的诱导率。试验建立了西瓜遗传转化和快速繁育的子叶再生系统。
The plantlet regengeration from cotyledon of 37 Xing and No 2 Zhongyu was studied. The results showed: the most suitable medium for bud induction from cotyledon of watermelon was MS+6-BA 5.0 mg·L^-1+IAA 0.5 mg·L^-1, and the rate of inducement arrived at 89.7%. Buds elongated at MS+KT 0.2 mg·L^-1. The optimum rooting media were MS+NAA 0.1 mg·L^-1, and the rate of inducement arrived at 88.2%. Compareing with the medium of MS+IAA 0.5 mg·L^-1. The system of genetic transformation and quick propagation for cotyledon of watermelon was established preliminarily.
