芥菜离体培养中玻璃化机理的研究 被引量:13

摘要 以宽帮青菜和红叶大头菜两个芥菜品种为试验材料 ,从形态解剖及生理生化方面探讨芥菜试管苗玻璃化的机理。结果表明 :宽帮青菜和红叶大头菜试管苗的玻璃化类型不同 ;宽帮青菜玻璃苗叶片解剖结构与正常苗有很大差异 ,红叶大头菜则无明显差异 ;玻璃苗叶片表面气孔器发育不完全 ,单位面积内气孔数减少 ;玻璃苗含水量增加 ,POD和PAL活性、木质素、叶绿素总量、叶绿素a、叶绿素b、RNA含量降低 ,但叶绿素a/叶绿素b、丙二醛和DNA含量、电导率、SOD活性几乎无变化。 Brassica juncea var. latipa cv.“Kuanbangqingcai” and B.Juncea var. megarrhiza cv.“Hongyedatoucai”were cultured in vitro to investigate the morphology, anatomy, physiology and biochemistry of vitrification. Great differences were observed between vitrified Kuanbangbaicai and Hongyedatoucai: anatomically, the leaves of vitrified shoots of Kuanbangbaicai were strikingly different from the normal shoots while no obvious difference existed between vitrified and normal shoots of Hongyedatoucai. The vitreous shoots of the former cultivar displayed tissue hyperhydrophy, were dark green in color and their leaves were thicker than the normal leaves; the vitreous shoots of the latter cultivar were translucent and light green. Stomata on leaf surface of the vitreous shoots developed abnormally and their number per unit area decreased. In vitreous shoots, water content increased, POD and PAL activity and the contents of total chlorophyll, chlorophyll a and chlorophyll b and RNA decreased, while the ratio of chlorophyll a/b, the contents of MDA and DNA, electrical conductivity and SOD acitivity remain almost unchanged.
