文心兰、蝴蝶兰类原球茎薄切片高频诱导PLBs 被引量:10

High Efficiency Regeneration of PLBs from Thin Cell Layers of PLBs of Oncidium and Phalaenopsis in Vitro

摘要 以文心兰、蝴蝶兰类原球茎薄切片为外植体,探讨了植物生长调节剂6-BA、Ad和NAA及其组合、蔗糖浓度及添加椰汁对类原球茎再生的影响。结果表明,特定浓度的3种植物生长调节剂的组合使用是PLBs高频发生的关键;培养基中添加椰汁能有效提高PLBs的诱导频率,培养基中高浓度蔗糖降低文心兰PLBs发生频率,但对蝴蝶兰没有影响;各处理诱导的PLBs在生长状况上差异不明显。适合两种兰花类原球茎薄切片高频诱导PLBs的固体培养基是1/2MS+4.0mg/L 6-BA+2.0mg/L Ad+1.0mg/LNAA+100~200m l/L椰汁+10~20g/L蔗糖+4.0g/L琼脂粉。 The thin cell layers (TCLs) of protocorm - like bodies (PLBs) of oncidium and phalaenopsis were used as explants cultured to the medium with different concentrations of plant growth regulators 6 - BA, Ad and NAA, and then different Goncentrations of coconut water and sucrose were added to the medium with given concentration of plant growth regulators. The aim was to investigate the effects of different concentrations of plant growth regula tors and their combinations, coconut water and sucrose on PLBs regeneration from the TCLs. The results showed that combinations of the given concentrations of three kinds of plant growth regulators were key factors in the PLBs regeneration from the TCLs in vitro. Additional coconut water in the given medium could raise the rate of PLBs induced from TCLs effectively. Higher concentrations of sucrose in the media reduced the efficieneies of PLBs formation of oncidium but no effects on phalaenopsis. The growth of PLBs induced from TCLs had no obvious differences among all treatments. The optimal medium for two kinds orchids of PLBs regeneration from TCLs was 1/2MS +4.0mg/L 6 - BA + 2.0mg/L Ad + 1.0mg/L NAA + 100 - 200ml/L coconut water + 10 - 20g/L sucrose + 4.0g/L agar powder.
