摘要
本文研究了春石斛‘森禾2006’的拟原球茎(PLBs)诱导、增殖、分化及壮苗和生根,建立了其组培快繁体系。结果表明:‘森禾2006’PLBs最适诱导培养基为1/2MS+TDZ 0.5 mg·L-1+水解酪蛋白2.0 g·L-1;PLBs增殖培养基为1/2MS+水解酪蛋白2.0 g·L-1;PLBs分化培养基为1/2MS+KT 1.0 mg·L-1+NAA 0.1 mg·L-1;壮苗生根培养基为MS+IBA 0.5 mg·L-1+NAA 0.1 mg·L-1+香蕉泥100.0 g·L-1。
To achieve the mass-scale multiplication of Dendrobium nobile 'Senhe 2006', the optimal media for the induction, multiplication and differentiation of protocorm-like bodies (PLBs), strengthening and rooting were studied and the rapid propagation system by tissue culture was established. The results showed that I/2MS+TDZ 0.5 mg.L%casein hydrolysate 2.0 g.L1 was optimal for PLB induction. 1/2MS+casein hydrolysate 2.0 g.L-1 was the most suitable for PLB multiplication. 1/2MS+KT 1.0 mg.L-1+NAA 0.1 mg-L-1 was the most efficient for PLB differentiation. The optimum medium for strengthening and rooting was MS+IBA 0.5 mg.L-1 +NAA 0.1 mg.L-l+banana homogenate 100.0 g.L-1.
