摘要
目的探讨转录因子Elf-1与凋亡抑制因子survivin在非小细胞肺癌中的表达及它们与临床病理因素之间的关系。方法收集72例非小细胞肺癌组织,应用组织芯片仪制备组织芯片,采用免疫组化PowerVisionTM-9000(PV-9000)法检测Elf-1、survivin的表达。结果在72例非小细胞肺癌组织中,Elf-1阳性染色定位于肺癌细胞核及胞质,阳性表达53例(73.61%),其表达强度与非小细胞肺癌的分化程度有关(χ2=14.849,P=0.021)。不同临床分期的非小细胞肺癌组织中Elf-1表达强度差异有统计学意义(χ2=24.127,P=0.000),Elf-1表达强度在淋巴结有转移组显著高于淋巴结无转移组(χ2=18.342,P=0.000);survivin阳性染色定位于肺癌细胞核及胞质,阳性表达44例(61.11%),其表达强度与非小细胞肺癌的分化程度有关(χ2=16.251,P=0.012),不同临床分期的非小细胞肺癌组织中sur-vivin表达强度差异有统计学意义(χ2=20.570,P=0.000),survivin表达强度在淋巴结有转移组显著高于淋巴结无转移组(χ2=8.723,P=0.033);非小细胞肺癌组织中Elf-1与survivin的表达强度呈极显著性正相关(τ=0.255,P<0.05)。结论Elf-1的表达强度与survivin的表达程度密切相关,均与肺癌细胞分化程度、临床分期及淋巴结转移有关,提示两者均可成为评估非小细胞肺癌生物学行为的重要指标。
Objective To study the expression of Elfq and relationship with survivin and biological behaveior in non-small cell lung cancer (NSCLC). Methods Tissue microarray from 72 cases of NSCLC was developed. The expression of Elf-1 and survivin were detected hy immunoistoehemistry staining with PV-9000 method. Results Elf-1 was mostly located in nucleus and in cytoplasm. The positive expression of Elf-1 was observed in 53 cases(73.61%), and its intensity of expression was related to the differentiation stages of the cancer (Х^2 = 14. 849, P = 0. 021), and significant differences were observed between different clinical stages (Х^2 = 24. 127,P = 0. 000). The intensity of Elf-1 expression was significantly higher in the group with lymph node metastasis than that without (Х^2 = 18. 342, P = 0. 000) ; Positive expression of survivin was also observed in 44 cases(61.11%), its intensity of expression was related to the differentiation stages of the cancer (Х^2 = 16. 251, P = 0. 012), and significant differences were noticed between different clinical stages (Х^2 = 20. 570, P = 0. 000). The intensity of survivin expression was significantly higher in the group with lymph node metastasis than that without (Х^2 = 8. 723, P = 0. 033) ; Positive correlation was found between the expression intensity of Elf-1 and survivin(r = 0. 255, P〈 0. 05). Conclusion The expression intensity of Elf-1 is closely correlated with survivin. The expression of Elf-1 and wurvivin is related to differentiation degree, clinical stage and lymph node metastasis of non- small-cell lung cancer. Elf-1 and survivin might serve as important parameters in evaluating biological behaviors of non small cell lung cancer.
