摘要
目的:了解动脉平滑肌细胞(ASMC)的蛋白激酶C(PKC)含量及亚型分布,以及与细胞增殖的关系。方法:采用点印迹技术及MTT方法,检测了人ASMC颗粒部分与胞浆部分PKC的6种亚型的含量和分布。结果:①人ASMC细胞中主要有PKCα、PKCβ、PKCζ3种亚型分布,并发现63%分布在胞浆中;②人ASMC细胞PKC总量随着佛波酯(PMA)、胰岛素浓度的增加而增加,人ASMC细胞增加数量也随着PMA、胰岛素的浓度增加而增加。结论:人ASMC细胞中以PKCα、PKCβ、PKCζ3型含量较多,胰岛素可能是通过激活PKC信号传导途径引起细胞的基因表达增高,导致ASMC细胞增殖。
Objective: To investigate the effects of insulin on distribution and content of protein kinaseC isoforms (PKCIs) in human arterial smooth muscle cells (ASMCs). Methods: Dot blot hybridization and MTT microenzymatic determination were used to detect the distribution of six subtypes of PKCIs and then to explore the relationship between the changes of subtypes and the proliferation of human ASMCs after the interference of insulin. Results: ①human ASMCs mainly contained PKCI subtypes alpha, beta and epsilon and 63% of them were distributed in cytoplasm; ②The total PKCIs in human ASMCs and ASMC count were increased along with the increase of concentration of insulin and PMA. Conclusion: Most of the PKCIs in ASMCs belong to subtypes alpha, beta and epsilon. Insulin induces the proliferation of ASMCs through activating the PKCI signal pathway.
