肾阳虚肺纤维化大鼠模型的研究 被引量:9

A New Rat Model with Renal-yang Asthenia and Pulmonary Fibrosis

摘要 目的:探索建立肾阳虚肺纤维化大鼠模型。方法:SD大鼠30只,随机分为生理盐水组(NS)、肺纤维化组(BLM)、肾阳虚纤维化组(RYA),每组10只,于一次性气管内滴入博莱霉素后,每日灌胃腺嘌呤,至第29天早上取标本处死大鼠,通过四诊表现、血肌酐(BUN)、尿素氮(Cr)、皮质醇(Cor)和肺肾病理改变及运用组织芯片结合胶原染色图象分析行肺胶原定量,鉴定肾阳虚肺纤维化大鼠模型。结果:肾阳虚肺纤维化组大鼠大体表现及肾病理改变与肾阳虚动物模型相似,肺病理改变与肺纤维化组相似,但肺胶原沉积程度(0.5413±0.2104)较肺纤维化组(0.9578±0.3756)轻(P<0.05),BUN、Cr(8.4872+2.8025和835.7333+377.2046)皆明显高于肺纤维化组(5.7863+1.3817和356.5400+300.5365)(P<0.05),Cor(14.1700+1.8999)则明显低于生理盐水组(20.4889+7.2648)和肺纤维化组(24.5040+9.4264)(P<0.05)。结论:肾阳虚肺纤维化大鼠同时具备了肾阳虚和肺纤维化的特征,说明模型新创成功,可作为进一步研究肾阳虚肺纤维化及温阳中药防治肺纤维化的动物模型。 Objective :To establish a new rat model with renal-yang asthenia and pulmonary fibrosis so as to study furtherly the pulmonary fibrosis with renal-yang asthenia syndrome and the effects of warming-yang Chinese traditional medi- cine on it. Methods :30 SD-rats were randomly divided into the NS-group (NS), the pulmonary-fibrosis group (BLM)and the renal-yang-asthenia-and-pulmonary-fibrosis group (RYA), each group 10 rats. Firstly pulmonary fibrosis rat models were established by blemycin-induced by way of once-dripping into trachea, in addition, pulmonary fibrosis with renal-yang asthenia rat models, as well as adding adenine by primed-stomach once each day ,until day 29 ,these rats were killed and the rat lung tissue and blood were acquired, by way of TCM-diagnosis associated with changes of BUN, Cr, Cor in the blood and pathologic changes of lungs and kidneys as well as tissue microarray associated with collagen-dye computer-picture analysis technique to quantify the collagen in the lung,the model were identified. Results:In RAY group, the rats resenble the renal-yang-asthenia-model rats in perfermance and renal pathological changes and the BLM rats in lung pathology, lighter than the BLM rats in collagen deposit(0. 5413 ±0. 2104 less than 0. 9578± 0. 3756, P 〈 0. 05), higher than the BLM rats in BUN and Cr( 8.4872 + 2. 8025 and 835.7333 ± 377.2046 higher than 5.7863 ±1. 3817 and 356. 5400 ± 300.5365 respectively,P 〈 0.05),lower than the NS rats and the BLM rats in Cor(14. 1700± 1.8999 lower than 20. 4889 ± 7. 2648 and 24. 5040 ± 9. 4264 respectively), P 〈 0. 05. Conclusion:the RYA rats with the characteristic of the BLM rats and the renal-yang-asthenia-model rats indicated that the RYA rat model were established successfully and could be used as the animal model for studying furtherly the pulmonary fibrosis with renal-yang asthenia and preventing and treating it with traditional Chinese medicine with warming-yang effect.
