肾阳虚小鼠肺及气道组织糖皮质激素受体β-防御素-2及其mRNA的表达 被引量:1

摘要 目的:探讨糖皮质激素受体(GR)、β-防御素-2(BD-2)及其mRNA在肾阳虚小鼠气道及肺组织中的表达及其意义。方法:45只清洁级KM小鼠随机分为对照组(NS)、肾阳虚组(RYA)及药物干预组(SIR),用腺嘌呤灌胃复制小鼠肾阳虚模型,通过免疫组化SP法测定肺及气道组织GR和BD-2的表达水平,原位杂交(ISH)法对GR和BD-2mRNA表达进行定位定量分析。结果:与正常组比较,肾阳虚组肺和气道组织GR及其mRNA表达下调(P<0.05),BD-2及其mRNA表达上调(P<0.05)伴肺组织炎症,与肾阳虚组比较,金匮肾气丸组GR及其mRNA表达上调(P<0.05),BD-2无统计学差异(P>0.05),但肺组织炎症程度减轻。结论:肾阳虚小鼠气道及肺组织GR及其mRNA表达下调,BD-2及其mRNA表达上调伴肺组织炎症,这是肾阳虚证本质研究在分子水平的进一步深入,在此状态下气道局部防御功能因BD-2等表达上调而增强,这可能是动用了气道-肺组织局部免疫储备功能的结果。 Objectives: To investigate the glucocorticoid receptor(GR) 、β-defensin-2and its m RNAs expression and value in the Airway Epithelial Cells and Lung Tissues of mice With Renalˉyang asthenia.Methods: 45 healthy KM mice,were randomly divided into the NS group(NS) 、the Renal-Yang-Asthenia group(RYA) and the kidney-qi Bolus group(SIR),the Renal-Yang-Asthenia mice models were established by adenine-inducing by way of primed-stomach once each day,until day 30,as well as adding the Six-Ingredient Rehmannia pill in the SIR;the mice were killed during 6-8am at day 31,the mice airway and lung tissue were acquired;and then the GR、BD-2 and its mRNAs were detected respectively by immunohistochemical way and in situ hybridization.Results: Comparing with the NS group,the GR and its mRNA’s expression were downregulated(P 〈 0.05),and the BD-2 and its mRNA’s were upregulated(P 〈0.05) in the airway Epithelial Cells and Lung Tissues with inflammation change in the Renal-yang asthenia mice;comparing the RYA,the GR and its mRNA’s expression were upregulated with inflammation decrease(P 〈 0.05),and no effects on the BD-2 and its mRNA(P 〉 0.05) in the airway Epithelial Cells and Lung Tissues in the SIR mice.Conclusion: GR and its mRNA,s expression downregulatin and the BD-2 and its mRNA upregulation in the airway Epithelial Cells and Lung Tissues with inflammation change in the Renal-yang asthenia mice,it were one of the molecular mechanism of the Renal-Yang-Asthenia Syndrome,and it might be utilized the airway-lung local immune store for future use that airway local defense increased due to BD-2 upregulation under the state of the Renal-Yang-Asthenia.
