摘要
目的探讨大承气汤(DD)在脂多糖(LPS)所致老年大鼠急性肺损伤(ALI)中的作用。方法将140只SD老年大鼠随机分为对照组、LPS组(经气管内滴注LPS复制ALI模型)、DD+LPS组和DD组。各组大鼠于给药后4或8h处死,测定肺系数;光镜观察肺组织形态学改变;化学法检测肺组织丙二醛(MDA)含量以及支气管肺泡灌洗液(BALF)的中性粒细胞(PMN)数目和蛋白含量的变化。各组再取7只动物,右心导管法监测并记录给药前及给药后2、4、6、8h时的平均肺动脉压(mPAP)。结果气管内滴注LPS可引起mPAP明显升高(P<0.01),肺组织明显的形态学改变;肺系数和肺组织MDA含量增加(P<0.01);BALF中PMN数目和蛋白含量增加(P<0.01)。给予DD后,mPAP明显降低,肺组织损伤减轻,肺系数和肺组织MDA含量下降(P<0.05),BALF中PMN数目和蛋白含量减少(P<0.01)。结论DD具有抗LPS所致老年大鼠ALI的作用。
Objective To explore the effect of Dachengqi decoction(DD)on acute lung injury(ALI)of aged rats induced by lipopolysaccharide(LPS)and its possible mechanisms.Methods140 SD aged rats were randomly divided into control,LPS,DD+LPS and DD groups,and were killed at 4 h or 8 h after administration.Lung indexes were tested;lung histomorphological changes were observed under microscope;and chemical method was used to measure the expressions of MDA in lung tissue and PMN in BALF.As for the other 7 rats of each group,right heart catheter method was used to monitor mean pulmonary arterial pressure(MPAP)before and 2,4,6,8 h after administration.ResultsMPAP was increased by instillation LPS into trachea,meanwhile the expressions of MDA in lung tissue and PMN in BALF were increased,and lung indexes were increased,and morphologic changes were appeared.With DD given,MPAP was decreased after given DD,at the same time,lung injury was lightened,lung indexes was decreased,and the expressions of MDA and PMN were reduced.ConclusionsDD can protect lung tissue of ALI rats induced by LPS.
