摘要
目的研究不同浓度，不同作用时间奥曲肽（Oct）对体外培养胃癌细胞MGC-803的作用及可能机制．方法采用MTT比色法检测Oct对体外培养胃癌细胞生长的抑制作用．用DNA断片法和流式细胞仪观察了Oct对胃癌细胞凋亡的影响，扫描电镜和透射电镜同时显示了凋亡细胞的形态学特征．结果MTT比色法测得Oct对胃癌细胞增殖有明显的抑制作用，呈剂量相关性Oct能有效地诱导人胃癌细胞MGC-803凋亡．经Oct处理12h～48h的胃癌细胞，DNA琼脂糖电泳呈现典型的梯形条带（Ladder），电镜和流式细胞仪（FcM）检测出现典型凋亡特征结论Oct对胃癌MGC-803细胞生长增殖有抑制作用；能有效地诱导人胃癌细胞MGC－803凋亡；
AIM To investipete the effects of octretide (Oct) on reproduction and apoptosis of human gastric cancer cell in vitro.METHODS By using MTT colorimetric assay, we oberved the antiprolifeation effect of different cancertions of Oct in vitro cultured human gastric cancer cell line MGC-803, Flowcytometry and DNA agarosey gel electrophores is revealed the characteristic apoptosis.RESULTS Oct inhibited MGC-803 cell apoptosis significantly correlative to drug concentration and time.
