摘要
通过构建一套整合载体,将来自嗜热脂肪芽孢杆菌的耐高温α-淀粉酶基因以及质粒pC194上的一段Cm抗性基因随机整合至枯草杆菌1A289染色体上,通过提高整合菌的抗氯霉素能力和反复进行整合,可提高耐高温α-淀粉酶基因在染色体上的拷贝数,最终耐高温α-淀粉酶基因在染色体上的拷贝数由整合初的一个增至七个以上。同时,在无选择压力的情况下。
By constructing a set of integration vectors,the thermostable α amylase gene from B.stearothermophilus and Cm resistance gene from pC194 were randomly integrated into the genome of B.subtilis 1A289.The number of the integrated thermostable α amylase gene copies was further increased by repeating the methed of random integration or by amplifying the gene using Cm selection,The copy number of the thermostable α amylase gene in genome increased from one to seven.Under the optimal conditions,the thermostable α amylase activity reached 220 u/ml without selection pressure.
