摘要
通过对一株高产耐高温α-淀粉酶的地衣芽孢杆菌摇瓶发酵工艺的优化,以及在摇瓶和20L发酵罐上进行的耐高温α-淀粉酶液态发酵工艺条件的试验研究,确定发酵培养基所采用的最佳氮源为豆饼水解液,最佳碳源为玉米粉液化液。正交试验结果表明:优化摇瓶发酵培养基配方为玉米液化液12.5%,豆饼粉5%,豆饼水解液2.5%,(NH4)2SO40.5%,摇瓶产酶活可达5000U/mL,比优化前提高了10%,20L发酵罐产酶活达到了12000U/mL。
The fermentation medium of Bacillus Licheniformis, which produces thermostable α-amylases were studied in shaking flask and 20 L-ventilating fermenter respectively. The suitable nitrogen source was the soybean flour acidolysis and the suitable carbon source was corn flour deliquesce. Anoptimum culture medium was obtained by orthogonal experiment, which is corn flour deliquesce 12. 5%, soybean flour 5% ,soybean flour acidolysis 2. 5%, (NH4)2SO4 0. 5%. thermostable α-amylases activity reaches 8 000 μ/mL in shaking flask,and 12 000 μ/mL in 20 L fermenter.
