摘要
用甲基葡萄糖及麦芽糖为底物,对50多株黑曲霉α-葡萄糖苷酶活性作了测定,结果表明各菌株对这二类底物之水解能力并不一致,甲基葡萄糖水解活性高的,其麦芽糖水解能力未必也高。通过TLC测定这些菌株生成异麦芽低聚糖之能力,结果表明甲基葡萄糖和麦芽糖为底物的水解能力并不能正确反映转苷反应的能力。酶活力高的,生成异麦芽低聚糖的能力未必比活性低的为强,造成上述结果的原因,或许是胞外酶中存在着仅有水解作用的α-葡萄糖苷酶和具有转苷酶活力的α-葡萄糖苷酶之故。试验表明臭曲霉sp.,黑曲霉CCRC31494,碳黑曲霉CCRC30414是作为生产葡萄糖苷转移酶的优良菌株,其中臭曲霉除生成异麦芽糖外,还生成显著量的潘糖。
The activity of α glucosidase of 50 strains of Asp.niger producing glucoamylase,acid protease and citric acid was determined by using methylglucoside and maltose as substrates.The results showed that the ability for hydrolysis of methylglucoside were not totally parallel with the ability for maltose.By using TLC technology,the results showed that neither the activity of hydrolysis of methylglucoside nor the activity of hydrolysis of maltose could reflect strain ability for the transgluosylation.The above results might be two kinds of extracellar α glucosidase containing in the extract of cultures:one only for the hydrolysis of substrate,and the other for both hydrolysis of substrate and transglucosylation.Furthermore,the experiments showed that Asp.foetidus sp.,A.niger CCRC 31494 and A.carbonarious CCRC 30414 were the potential strains for producing transglucosidase.
