摘要
目的建立快速测定青稞秸秆中β-葡聚糖含量的方法。方法采用分光光度法,刚果红为显色剂,测定最佳条件为:溶液pH为8.0、反应温度为20℃、反应时间为30min、最大吸收波长为550nm。结果β-葡聚糖测定的线性范围为0~10μg/mL(r=0.9996),平均回收率为99.31%,RSD=1.29%。结论该测定方法简便、准确、重复性好,适用于青稞秸秆及其它植物中β-葡聚糖含量的测定。
Objective To establish a method to determine the β-glucan content in the Hullessbarley Straw stalk.Method Spectropho-tometry was used,Congo red was colorant.The optimal determination conditions were as follows:the acidity of solution pH was 8.0,reac-tion time was at 30 min,reaction temperature at 20 ℃ and the maximum absorption wavelength at 550 nm.Results The β-glucan had a good linearity in the range of 0~10 μg/mL(r =0.999 6) and the average recovery was 99.31 % with RSD of 1.29%.Conclusion The method was simple ,accurate and reproducible. It is suitable for the determination of the β -glucan content in the Hullessbarley Straw stalk and other plants.
