摘要
为探讨白细胞介素1对肾小球系膜细胞作用的细胞内信号转导机制,采用3H胸腺嘧啶参入、免疫沉淀及激酶磷酸化反应等方法,检测了白细胞介素1对分裂原活化蛋白激酶不同亚类的作用,并与血小板衍生生长因子的作用进行比较。结果发现,与血小板衍生生长因子的作用相反,白细胞介素1可在短时间内激活cjun氨基末端激酶,但对p42/44分裂原活化蛋白激酶无刺激作用,表明白细胞介素1在肾小球系膜细胞的作用可通过cjun氨基末端激酶介导。
おhe signalling pathways by which IL1 exerts its effects in renal mesangial of subfamily of mitogenactivited protein kinases(MAPKs) stimulated by IL1.DNA synthesis in MsC was detected by 3HThymedine uptake.p42/44 MAPK activity was measured by phosphorylation of myelin basic protein.Cjun aminoterminal kinase(JNK)activity was measured by phosphorylation of GSTcjun immobilized to glutathioneagarose.The results revealed that IL1β seems to be a very weak stimulatory cytokine for DNA synthesis in MsC.IL1β(10 ng/ml)stimulated JNK activity by 2~3 fold in 15 min and return to normal by 1~2 hour.However,IL1β had no affect on p42/44 MAPK activity. In contrast to IL1β,polypeptide pletletderived growth factor(PDGF)rapidly activated p42/44 MAPK,but no effect on JNK from 15 min to 2 hours. These results suggested that effects of IL1 in MsC may be mediated through activation of JNK.The different signal pathway may play the distinct role on MsC proliferation and differentiation.
