摘要
将雄性纯种新西兰家兔36只随机分为三组,喂饲胆固醇形成实验性高胆固醇血症,部分加用麝香保心丸喂养;同时采用定量RT-PCR等技术,观察高脂血症对动脉壁一氧化氮系统代谢的影响,以及麝香保心丸的保护作用.结果表明:高脂饮食可造成动脉一氧化氮代谢的紊乱,降低动脉壁内皮型一氧化氮合酶(eNOS)基因的表达(P<O.05)、减少动脉壁一氧化氮合酶(NOS)的活力、降低血清一氧化氮代谢产物(NOP)的水平(P<0.05).麝香保心丸与高脂血症对照组相比,可显著增强动脉NOS的活力、提高血浆一氧化氮代谢产物的水平(P<O.05).
Purpose:This study was designed to determine the effect of hyperlipidemia and the outcome of intervention with Heart-protecting musk pill (SXBXW) on nitric oxiole(NO) metabolism of arterial wall. Methods :Thirty-six male Newzealand rabbits were randomized into three groups: control group(C),fed with common diet for 8 weeks. hypercholesterolemia group(HC) ,fed with' 1% cholestsrol and 3% pig oil diet for 8 weeks and drug intervention group (SX) in which the rabbits were fed with both cholesterol-rich diet and SXBXW(6 # /day during the last 2 weeks). The level of endothelial nitric oxide synthase (eNOS) mRNA was detected by quantitative reverse transec ription polymerase chain reaction(RT-PCR). Nitric oxide synthase(NOS) activity of arterial wall was measured in three groups at the 8th week. Serum nitric oxide production(NOP) was assessed by modified Greiss method at the 6th and 8th week. Results ; The data showed that hyperlipidemia caused decrease in eNOS mRNA levels and down-regulate NOS activity. decrease the levels of serum NOP(Compared with C group (P<0. 05). But SXBXW partly reverse the impaire-ment of NO system. It had a tendency of increaseing eNOS mRNA levels (compared with HC group, 0. l>P>0. 05) and markedly elevated the NOS activity and the concentration of NOP (compared with HC group (P<0. 05). Conclusion: The results suggest that the dysfunction of NO metabolites may play an important role in the development of atherosclerosis; SXBXW can decrease, in partly, this impairement.
