三磷酸腺苷法肿瘤药敏在口腔颊黏膜鳞癌化疗中的实验研究 被引量:1

摘要 目的:采用三磷酸腺苷法(ATP-TCA)对口腔颊黏膜鳞癌组织标本进行肿瘤药敏检测,为口腔颊黏膜鳞癌的个体化化疗选择提供依据。方法:45例口腔颊黏膜鳞癌新鲜标本行肿瘤细胞分离,6种化疗药物分别和癌细胞原代培养后,进行ATP-TCA检测。结果:所有标本均顺利完成检测,可评估率为100%,单药强敏感率为吡柔比星44.44%、替尼泊苷35.56%、表柔比星31.11%、顺铂22.22%、氟尿嘧啶20.00%、紫杉醇13.33%,不同药物对口腔颊黏膜鳞癌的强敏感程度有差异(P=0.012),该组不同个体肿瘤细胞对不同药物的敏感性程度差异具有显著统计学意义(P=0.0006)。结论:ATP-TCA可为口腔颊黏膜鳞癌的临床个体化化疗提供重要的指导作用。 Objective:To test the chemosensitivity of patients with squamous carcinoma of buccal mucosa using adenosine triphosphate based tumor chemosensitivity assay (ATP-TCA)in order to provide a basis for individualized chemotherapy. Method: ATP-TCA was used to determine the effect of 6 chemotherapy drugs on surgical specimens from 45 patients with squamous carcinoma of buccal mucosa. Results: All the 45 specimens were successfully tested and the rate of evaluation was 100%. There were considerable differences in chemosensitivity between individuals. The most active single drug was pirarubicin (44. 44% ). The sensitivity of other five drugs in order of decreasing sensitivity were teniposide ( 35.56% ), epirubicin ( 31.11% ), cisplatin ( 22.22% ), 5-Fu ( 20.00% ) and paclitaxel ( 13.33 % ). There was individual difference in the in vitro chemosensitivity of squamous carcinoma of buccal mucosa and the difference had statistical significance (P = 0.0006 ). Conclusion:The use of the ATP-TCA may provide a method for selecting appropriate anti-cancer drugs for squamous carcinoma of buccal mucosa .
