用MTT法进行白血病体外药物敏感性检测的临床意义 被引量:10

摘要 目的探讨使用MTT比色法进行白血病体外药物敏感性检测的可行性,为MTT法作为肿瘤药敏检测技术应用于临床提供理论依据。方法采用MTT比色法测定47例血液病患者外周血和骨髓白血病细胞对14种临床常用化疗药物的敏感性,并与化疗药物的实际疗效作对照。结果47例患者中,体外敏感/体内敏感(S/S)30例、体外敏感/体内耐药(S/R)7例、体外耐药/体内耐药(R/R)9例、体外耐药/体内敏感(R/S)例;体外药敏与体内疗效的总符合率为82.98%,阳性符合率为81.09%,阴性符合率90%。敏感度96.77%,特异度56.25%。结论采用MTT法预测化疗药物的敏感性,临床相关性好。 Objective To investigate the feasibility of the application of MTT assay in the in vitro leukemia chemosensitivity test, in attempt to provide theoretical basis for its clinic application. Methods Peripheral blood cells and bone marrow leukemia cells collected from 47 patients were detected by MTT colorimetric assay in light of their sensitivity to 14 drugs. The relationships between the findings and the practicalities of ehemotherapeutics were analyzed. Results Among 47 patients,30 cases were S/S,7 cases S/R,9 cases R/R and 1 case R/S (S: sensitivity, R: resistance, in vitro/in vivo). The rates of general coincidence, positive coincidence, negative coincidence, sensitivity, specificity were 82. 98%, 81. 09%, 90%, 96.77 %, and 56.25%, respectively. Conclusion The employment of MTT assay as an approach to estimating the sensitivity of chemotherapeutics has a good correlation to clinical practice, which may provide guidance for clinical leukemia chemotherapy.
