摘要
目的:观察内毒素对大鼠肺泡Ⅱ型上皮细胞(typeⅡpenumonocyte)水通道蛋白5(aquaporin-5)的影响。方法:对大鼠肺泡Ⅱ型上皮细胞进行分离、纯化,用1μg/mL的脂多糖(lipopolysaccharide,LPS)刺激肺泡Ⅱ型上皮细胞4h,并将肺泡Ⅱ型上皮细胞随机分为正常对照组和LPS组。分别采用逆转录聚合酶链反应法(RT-PCR)和免疫组织化学法(IHC)检测肺泡Ⅱ型上皮细胞中水通道蛋白5的表达。结果:RT-PCR结果显示LPS刺激肺泡Ⅱ型上皮细胞后水通道蛋白5的信使核糖核酸(mRNA)水平明显低于正常对照组(P<0.01)。免疫组织化学法显示大鼠肺泡Ⅱ型上皮细胞上水通道蛋白5呈阳性染色,且LPS刺激后肺泡Ⅱ型上皮细胞上水通道蛋白5阳性细胞率显著低于正常对照组(P<0.01)。结论:大鼠肺泡Ⅱ型上皮细胞上存在水通道蛋白5,且水通道蛋白5表达减少可能与LPS引起的肺水肿有关。
Objective To investigate the expression of aquaporin(AQP)-5 in the rat type Ⅱ penumonocyte (AT Ⅱ ) treated by lipopolysaccharide (LPS), and to investigate the action mechanisms of cleating alveolar edema fluid in AT Ⅱ. Methods The rat AT Ⅱ were isolated and puritfied for the cellular uhrastrueture study. The AT Ⅱ were treated by 1 μg/mL LPS and divided randomly into two groups: the control group and LPS group. The expression of AQP5 in AT Ⅱ was detected by reverse transcription-polymerase chain reaction (RT-PCR) and immunoeytochemistry (IHC) assays 4 h later. Results The result of RT-PCR showed that the AQP5 mRNA expression was significantly decreased in the LPS group (P 〈 0.01 vs control group). The result of IHC showed that AQP5 was expressed in AT Ⅱ , and the AQP5 protein levels was significantly decreased in the LPS group compared with that of the control group (P 〈 0.01 ). Conclusion The above results suggest that the AQP5 can be expressed in the rat AT Ⅱ , and the down-regulation of the AQP5 expression may be associated with the LPS-induced lung edema.
