摘要
目的探讨甘氨酸对肝纤维化及肝硬化大鼠肝组织肝细胞生长因子(HGF)表达的影响。方法将40只雄性Wis-tar大鼠随机分为5组:正常对照组(C组),肝纤维化(HF)模型组,肝硬化(HC)模型组,肝纤维化甘氨酸干预组(HF+G)及肝硬化甘氨酸干预组(HC+G)。观察外周血浆内毒素(endotoxin),丙氨酸氨基转移酶(ALT)和血清肿瘤坏死因子α-(TNFα-)水平,免疫组化染色分析肝组织肝细胞生长因子(HGF)的表达情况。结果血浆endotoxin、ALT及血清TNFα-水平在HF及HC组明显高于C组(P<0.01),而在HF+G及HC+G组分别低于HF及HC组(P<0.05)。HGF在HF及HC组的表达均显著高于C组(P<0.01,P<0.05),在HC组的表达低于HF组(P<0.05),而在HF+G及HC+G组分别高于HF及HC组(P<0.05)。结论甘氨酸可减轻肝纤维化及肝硬化大鼠的肝损伤,这可能部分是通过上调肝组织HGF的表达实现的。
Objective To explore the effects of glycine on the expression of hepatocyte growth factor（HGF）in liver tissue of rats with hepatic fibrosis and cirrhosis.Methods Forty Wistar rats were randomly divided into five groups：control group,hepatic fibrosis（HF）group,hepatic cirrhosis（HC）group,hepatic fibrosis treated with glycine group（HF＋G）and cirrhosis treated with glycine group（HC＋G）.The levels of plasma endotoxin,alanine aminotransferase（ALT）,serum tumor necrosis factor-α（TNF-α）were detected.The expression of HGF was determined with immunohistochemistry.Results The levels of plasma endotoxin,ALT and serum TNF-α were significantly higher in HF group and HC group than in control group（P〈0.01）,but they were lower in HF＋G group and HC＋G group than in HF group and HC group,respectively（P〈0.05）.The expression of HGF was significantly higher in HF group and HC group than in control group（P〈0.01,P〈0.05）,but it was lower in HC group than in HF group（P〈0.05）.The expression of HGF was also higher in HF＋G group and HC＋G group than in HF group and HC group,respectively（P〈0.05）. Conclusion Glycine could lighten liver injuries of rats with hepatic fibrosis or cirrhosis partly by inducing the expression of HGF in liver tissues.
