骨髓间充质干细胞向视网膜神经节样细胞的诱导分化 被引量:5

摘要 目的探讨视网膜条件分化液诱导骨髓间充质干细胞(bone mesenchymal stem cells,BMSCs)向视网膜神经节样细胞定向分化的作用。方法采用PCR技术、流式细胞术、免疫荧光法观察视网膜条件分化液对BMSCs分化的影响。结果视网膜条件分化液诱导BMSCs3d后,Nestin阳性细胞比率为(30.9±7.7)%。诱导后第7天,免疫荧光结果显示Neurofilament的阳性细胞比率为(23±4)%,β-tubulin Ⅲ的阳性细胞比率为(18±3)%。RT-PCR结果也显示上述神经元或神经节细胞标志物的表达阳性。结论视网膜条件分化液对BMSCs向视网膜神经节样细胞定向分化具有促进作用。 Objeetive To observe the effect of supernatant from retinal neurons and glia cell on the differentiation of bone mesenchymal stem cells into retinal ganglionlike cells. Methods Effects of the supernatant of retinal neurons and glia cells on differentiation of bone marrow mesenchymal stem cells were observed by PCR, flow cytometry and immunofluorescence, Results After the BMSCs were induced by supernatant of retinal neurons and glia cells, the cells ratio were immunoreactive for neuronal specific marker nestin on day 3, which was ( 30.9 ±7.7 ) %. On day 7, positive cells ratio of neurofilaments were (23 ±4)% and positive cells ratio of β-tubulin Ⅲ were (18 ± 3)%. RT-PCR results showed that the above retinal ganglion cells marker were also positive. Conclusion Supernatant of retinal neurons and glia cells can promote the differentiation of bone mesenchymal stem cells into retinal ganglion-like cells.
