摘要
目的 :探讨聚合酶链反应 (PCR)技术对角膜内皮炎的病原学诊断价值。方法 :用PCR技术对角膜内皮炎房水及泪液中的单纯疱疹病毒Ⅰ型DNA进行扩增 ,并用琼脂糖凝胶电泳对PCR产物进行检测 ;同时以老年性白内障房水及泪液为对照组。结果 :16例角膜内皮炎房水中 11例阳性 ,阳性率为 68 75 % ,2 0例对照组房水中无 1例阳性 ,二者有显著性差异 (P <0 0 5 ) ;16例角膜内皮炎泪液中 3例阳性 ,阳性率为 18 75 % ,2 0例对照组泪液中 1例阳性 ,阳性率为 5 % ,二者无显著性差异 (P >0 0 5 ) ;角膜内皮炎房水检则阳性率 (68 75 % )与泪液检测阳性率 (18 75 % )有显著性差异 (P <0 0 5 )。结论 :用PCR技术检测角膜内皮炎房水中单疱病毒DNA可以对角膜内皮炎做出病原学诊断 ,并可进一步指导临床治疗。
Objective:To explore the gene diagnosis of corneal endotheliitis by polymerase chain reaction (PCR). Methods:Herpes Simplex Virus-I (HSV-I) DNA in aqueous humors and tears of the patients group with corneal endotheliitis (16 eyes of 11 cases)and the control group with cataract were detected by PCR and agarose gel electrophoresis. Results:68 75% of the endotheliitis eyes were positive for HSV-I DNA in aqueous humors,but no eyes was positive in the control group,the difference between the two groups was significant (P<0 05).The positive rates in tears between the two groups were not significantly different.The positive rates,however,in aqueous humors was significantly higher than that in tears(18 75%) in endotheliitis group. Conclusions:Etiologic diagnosis of corneal endotheliitis could be made with the detection of HSV-I DNA in aqueous humor by PCR and agarose gel electrophoresis,it is helpful for clinical treatment.
