摘要 目的探讨自体造血干细胞动员是否可使成体造血干细胞(HSCs)环境诱导分化,修复受损的视网膜微血管内皮细胞,使血-视网膜屏障得到功能性重建。方法糖尿病小鼠和正常小鼠各分为干细胞动员组和生理盐水对照组。流式细胞术以CD34-/low和sca1+为标记鉴定外周血HSCs。小鼠视网膜病理切片行苏木精-伊红染色观察和VEGF免疫组织化学分析,伊文思蓝定量检测血-视网膜屏障的破坏程度。结果自体干细胞动员使糖尿病小鼠视网膜的VEGF表达量明显下降。糖尿病小鼠经干细胞动员后清蛋白的渗出量明显下降(P=0.033)。结论自体HSCs动员可以通过外周血干细胞倍增使血-视网膜屏障得到一定程度的功能性修复。 Objective The break of blood-retina barrier is an important factor in aggravation of diabetic retinopathy. The purpose of this study was to evaluate hematopoietic stem cells (HSCs) multiplication by autologous hematopoietic stem cell mobilization and determine whether hematopoietic stem cell mobilization can ameliorates the break of blood-retina barrier in diabetic mice. Methods Twenty-two 15-week old KK mouse mice and 22 normal C57BL/6 mice were divided into four groups: normal mice control group,normal treated group, diabetic mice control group and diabetic treated group. 200 μg/(kg · d) of murine SCF and 50 μg/(kg · d) of rhG-CSF were injected subdermally for 5 days in the normal treated group and diabetic treated group,and equal volume of normal saline was used in the same way in normal and diabetic control groups. Flow cytometry, immunohistochemistry method were used to analyze the changes associated with autologous hematopoietic stem cell mobilization. Evans blue quantitative test was applied to measure the breakdown level of blood-retina barrier. HSCs were marked by CD34-/low and Sea-1 ^+ in this study. Results Vascular epithelial growth factor(VEGF) expression was markedly decreased in normal and diabetic treated groups. The increase of retinal thickness,the higher expression of VEGF in the ganglion cell layer and inner nuclear layer of retina were seen in the diabetic control group. The albumin leakage was slightly increased in the mice in normal treated group but no statistically significant difference in comparison with normal control mice(P = O. 186). HSCs could significantly reduce the leakage of albumin in the retina of diabetic mice ( P = 0.033 ). Conclusion Autologous HSCs mobilization can be therapeutic option in vascular reconstruction and functional restoration of blood-retina barrier in diabetic mice.
