摘要 目的研究校正PCR在性别鉴定中的可应用性。方法采用突变敏感性"分子开关"介导的PCR针对Y染色体特定序列进行识别。实验包括三个方面:采用普通PCR,针对Y染色体M45(rs2032631)位点设计特异性引物结合突变敏感性分子开关,对已知性别的样本进行检测;采用实时定量PCR和突变敏感性"分子开关",以及Y染色体特异性引物Y02对已知性别的样本进行检测;采用Y染色体特异性引物Y02对未知性别的男女模板进行检测。结果本实验对所采用的小数量样本,在已知性别组与未知性别组的性别鉴定上均达到检测性别与实际性别完全相符,预示着该方法在性别鉴定上的巨大潜在应用价值。 [Objective] The object of this experiment is to study the application of proofreading PCR in gender identification. [Methods] Three different experiments were performed: DNA sample with known gender identification was tested by regular PCR with primers targeting M45 SNP site in Y chromosome; DNA sample with known gender identification was tested by real time PCR and the mutation sensitive ON/OFF switch with Y chromosome specific primer Y02; Samples of unknown sex identification were determined with Y chromosome specific primer Y02. [Results] Based on the small number of DNA samples used in this study, the sex identification results of samples of known and unknown are both 100% correct. The data indicate that the mutation sensitive on/off switch has potential application in gender identification.
