摘要
目的探讨阿尼西坦对血管性痴呆(VD)大鼠海马脑源性神经营养因子(BDNF)表达的影响。方法采用Pulsinelli四血管阻塞改良法建立VD大鼠模型,造模后给予阿尼西坦;用水迷宫检测VD大鼠学习记忆能力,免疫组织化学SABC法检测BDNF阳性细胞。结果阿尼西坦治疗后VD大鼠学习记忆能力明显提高,海马BDNF免疫阳性神经元明显增多(P<0.05)。结论阿尼西坦可改善VD大鼠学习记忆能力,其机制可能与上调BDNF表达、保护缺血后海马神经元有关。
Objective To investigate the effect of Aniracelam (Ani) on the expression of brain-derived neurotrophic factor (BDNF) of hippocampal in vascular dementia ( VD ) rats. Methods VD rats models were established by employing improved method of Pulsinelli four-vessel occlusion. After estahlishing the models, the rats were treated with Ani. The abilities of learning and memory of the VD rats were assessed by MoITiS Water Maze Behavior Test. The numbers of BDNF positive neurons in the hippocampal were detected by immunohisto- chemistry SABC method. Results The abilities of learning and memory of the VD rats were improved and the expression of BDNF in hippocampus was increased after treated by Ani( P 〈 0. 05 ). Conclusions By increasing the expression of BDNF and protecting neurons in the hippocampus following ischenfia, Ani may improve the abilities of learning and memory of the VD rats.
