中药关节康对骨性关节炎软骨TGF-β1、IL-1β表达的影响 被引量:9

Effect of Guanjiekang on Expressions of IL-1β and TGF-β1 in Osteoarthritis Cartilage

摘要 目的:观察复方中药关节康对骨性关节炎软骨TGF-β1、IL-1β表达的影响,探讨其疗效机制。方法:通过左膝关节腔内注入木瓜蛋白酶造成骨性关节炎模型,36只新西兰兔随机分为6组:A组(正常组)、B组(造模组)、C组(空白对照组)、D组(关节康组)、E组(西乐葆组)和F组(壮骨关节丸组)。木瓜蛋白酶注射结束后2周D、E、F组分别予以关节康、西乐葆、壮骨关节丸治疗4周,取左膝关节软骨行IL-1β、TGF-β1免疫组化染色。结果:关节康组提高关节软骨TGF-β1表达的效果最明显,而西乐葆组降低关节软骨IL-1β表达的效果最明显。结论:关节康治疗膝骨性关节炎的疗效机理可能在提高关节软骨的TGF-β1含量,降低IL-1β含量,从而直接或间接促进了损伤关节软骨的修复。 Objective:To observe the effect of Guanjiekang (composed of Radix Rehmanniae Preparata, Radix Achyranthis Bidentatae, etc) on the expressions of IL-1β and TGF-β1 in osteoarthritie cartilage and to explore its possible mechanisms. Methods: Osteoarthritis models of News Zealand rabbits were established via left articular genu injection of papain. Thirty --six rabbits were randomly divided into normal group A, model group B, blank control group C, Guanjiekang group D, celecoxib group E and Zhuanggu Guanjie Pills group F. Two weeks after injected of papain, rabbits in group D, E and F were treated with Guanjiekang, celecoxib and Zhuanggu Guanjie Pills respectively for 4 weeks. Left knees were collected to measure the expressions of IL-1β and TGF-β1 with immunohistochemistry assay. Results:The expression of TGF-β1 significantly increased in Guanjiekang group, while the expression of IL-1β decreased the most in celecoxib group. Conclusion: Guanjiekang might promote the healing of osteoarthritis directly or indirectly via enhancing the expression of TGF-β1 and reducing the expression of IL-1β in cartilage.
