关节牵伸对兔膝骨关节炎软骨中IL-1及MMP-13表达的影响 被引量:2

Changes of interleukin-1 and matrix metalloproteinase-13 expression in Rabbit osteoarthritis cartilage after joint distraction

摘要 目的:应用环形外固定对兔膝骨性关节炎关节进行有限持续关节牵伸,观察一段时间后其对关节软骨组织形态学和白细胞介素-1(IL-1)及基质金属蛋白酶-13(MMP-13)表达的影响。方法2014年3至8月,将18只新西兰大白兔随机分为对照组、模型组及牵伸治疗组,各6只。模型组及治疗组于膝关节腔内注射木瓜蛋白酶建立兔膝骨性关节炎模型。造模成功后,治疗组手术安装特制外固定支架行4周持续关节牵伸。牵伸结束后观察各组家兔软骨组织形态学的变化及软骨中IL-1和MMP-13的蛋白表达变化。结果病理学检查显示模型组软骨病理损害程度重于对照组(P〈0.05),软骨组织中IL-1及MMP-13的表达也明显高于对照组(P〈0.05)。而经关节牵伸治疗后,软骨的病理损害程度较模型组减轻(P〈0.01),软骨组织中IL-1及MMP-13的表达也较模型组显著减少。结论应用环形外固定对骨性关节炎关节进行持续稳定的关节牵伸可促进软骨修复,抑制软骨组织中IL-1及MMP-13的表达。 Objective To investigate changes of IL-1 and MMP-13 expression level in rabbit articular cartilage and evaluate the pathological changes of cartilage histology after appling annular external fixator to draft knee osteoarthritis in limited continuous distraction in rabbit models. Methods 18 New Zealand rabbits were randomly divided into control group,model group and drafting group. OA model was established by injecting papain in the knees of rabbits in group B and C. After the modeling,the therapy group carried out surgery to install a special external fixator,which could provide drawing intensity during four weeks treatment. At the end of the experiment,pathological changes of cartilage and IL-1 and MMP-13 protein expression were observed in each group. Results Pathological examination revealed the degree of cartilage damage in model group was higher than the control group (P〈0.05),the expression of IL-1 and MMP-13 in cartilage tissue were also significantly higher (P〈0.05). After joint distraction, pathological damage of cartilage reduced compared to the model group(P〈0.01),the expression of IL-1 and MMP-13 also significantly decreased than the model group. Conclusion Results of joint distraction in osteoarthritis suggest that the therapy promote cartilage restoration and inhibit the expression of IL-1 and MMP-13 in cartilage tissue.
