淫羊藿对颈椎病模型大鼠退变颈椎骨组织钙磷代谢及血清中IL-lβ、IL-6、TNF-α含量变化的实验研究 被引量:9

Effects of Epimedium on Calcium and Phosphorus Metabolism in Degenerated Cervical Bone and Serum IL-lβ,IL-6 and TNF-a in Cervical Spondylosis in Rats

摘要 目的:研究淫羊藿对颈椎病模型大鼠退变颈椎骨组织中Ca、P代谢及血清中IL-1β、IL-6、TNF-α含量的影响,探讨淫羊藿防治颈椎病的机制。方法:30只SD大鼠,随机分为模型组20只和假手术对照组10只,采用切除动物C2~C7棘上韧带、棘间韧带和分离椎旁两侧肌肉,建立颈椎退变动物模型。在1月、3月两个时间点拍摄颈椎X线片,证实模型成立后,将模型组分为淫羊藿模型组(A组)和生理盐水模型组(B组)各10只,分别喂服淫羊藿水提液和生理盐水,假手术对照组(C组)喂服生理盐水,喂服2个月取大鼠的颈椎骨组织和血清,用全自动生化分析仪测定各组颈椎骨组织中Ca、P、ALP含量和发射免疫法测定血清样本中IL-lβ、IL-6、TNF-a指标。结果:①B组颈椎骨组织中Ca、P的水平低于其他两组(P<0.01)。A组Ca、ALP的水平高于其他两组(P<0.01),P的水平高于B组(P<0.01)。②B组血清中IL-1β、IL-6和TNF-α的含量高于C组(P<0.05、P<0.01),A组血清中IL-1β、IL-6和TNF-α的含量显著的低于B组(P<0.01)。结论::①颈椎Ca、P含量的减低,是颈椎退变的病理结果。淫羊藿可提高退变颈椎骨组织Ca、P含量,抑制颈椎退变。②淫羊藿能下调退变颈椎血清中IL-1β、IL-6和TNF-α炎性细胞因子的含量,具有阻止炎性反应和抑制颈椎退变的作用。 Objective:The study was to investigate the effect of Epimedium on calcium (Ca) and phosphorus (P) metabolism in degenerated cervical bone and serum IL-1β, IL-6 and TNF-α in cervical spondylosis (CS) rats so as to explore mechanisms of Epimediurn in the prevention and treatment of CS. Methods:Thirty SD rats were randomly divided into model group (20 rats) and sham operated group (group C, 10 rats). The models were established via cutting off C2--C7 supraspinal and interspinal ligaments and seperated the muscle groups beside cervical vertebra. Cervical vertebrae X--ray was used to check whether the models were successfully set up at 1st and 3rd months. After the models were established successfully, the rats with CS were randomly divided into Epimedium group (group A) and saline model group (group B). Group A were treated with Epimedium, while group B and group C were administered with saline every day for 2 months. The rats were killed to collect cervical bone and serum after the treatment. Automatic biochemistry analyzer was used to detect the content of Ca, P and ALP, and radioimmunity assay was applied to measure serum IL-1β, IL-6 and TNF-α. Results:(1)Ca and P levels in group B were lower than those in the other groups (P〈0.01). Ca and ALP levels in group A were significantly higher than those in the other groups (P〈0.01), while P content was significantly higher than that in group B (P〈0.01). (2) Serum IL-1β, IL-6 and TNF-α in group B were significantly higher than those in group C (P〈0.05 ,P〈0.01). Serum IL- 1β, IL-6 and TNF-α in group A were lower than those in group B (P〈0.01). Conclusion:(1)The reduction of cervical vertebral calcium and phosphorus was the pathological results after degenerated cervical vertebrae. Epimedium can improve the contents of calcium and phosphorus in degenerated cervical vertebra as well as prevent the degeneration of cervical bone. (2) Epimedium can decrease serum IL1-β, IL-6 and TNF-α in degenerated cervical vertebra, playing the functions of anti--inflammation and inhibiting degeneration of cervical vertebra.
