A Study on the Corncob Enzymatic Saccharification Conditions with Xylanase

摘要 为获得木聚糖酶酶解玉米芯的最适工艺条件,通过单因素试验,利用木聚糖酶对经2 ml/LHCl预处理的玉米芯在不同条件下进行酶解糖化,研究结果表明:在pH5.2的Na2HPO4-柠檬酸缓冲体系中、木聚糖酶用量150 IU/g玉米芯、酶解温度45℃、摇床转速120 r/min、酶解时间12 h,酶解液溶出还原糖量、总糖量分别达到42.35%和46.59%,其中预处理溶出还原糖量、总糖量分别为21.69%和30.00%;且在最适酶解工艺条件下,可获得较好的酶解效果。 To obtained the optimum processing condition for enzymatic saccharfication of corncob with xylanase, with thexylanase enzymatic saccharification of corncob, which was pretreated with 0.2% HCl solution, was systematically studied by single factor experiment in different conditions. In the buffer solution of Na2 HPO4 -Citric acid of pH 5.2, xylanase amount, 150 IU/g corncob, agitation rate 180 rpm, reaction temperature 45 ℃, hydrolysis time 12 h. Under above conditions the yield of reducing sugar and total sugar could be up to 42.35% and 46.59% (based on corncob in raw material ) respectively,The yield of reducing sugar and total sugar of pretreatment were 21.14% ,29.37%. respectively, Under the optimun process conditions, the fairly good effect of enzymatic saccharfication may gaied.
