摘要
采用聚乙二醇6000修饰物混合吐温80-硫酸铵盐-水液-固体系分离血红蛋白.研究了此萃取体系中修饰物的含量、成相聚合物吐温80的浓度、盐的浓度、体系酸度等因素对血红蛋白在液固两相中分配的影响,并得到最佳分离条件.体系中修饰物含量为2%(质量密度),吐温80含量为4.5%(体积百分数),酸度为pH=7.95,盐浓度为2mol/L.一次固相收得率达到96.5%.
The method of separation and purification of hemoglobin by using inorganic solvent liguid-solid extraction system of PEG-Met-Cu-tween80-(NH4)2SO4 was studied. The distribution of hemoglobin in the liquidsolid two phases can be changed by various factors such as concentrations of modifier, ammonium sulfate, and tween80, the value of pH and so on. Optimum extraction of hemoglobin was achieved at pH 7.95, PEG-Met-Cu modifier 2% (mass density ), Tween80 4.5% (volume fraction) (NH4)2SO42 mol/L.
