摘要
研究虎杖疏松愈伤组织诱导的最佳因素组合。通过单因素和正交试验研究不同外植体、不同浓度的非激素水平和激素水平对虎杖愈伤组织诱导率、疏松程度以及增殖率的影响。结果表明:以叶柄为外植体更容易诱导出疏松的愈伤组织,虎杖愈伤组织诱导的最佳非激素因素为pH值5.8,蔗糖5 g/L,水解酪蛋白300 mg/L,肌醇10 mg/L,继代增殖最佳的激素组合2,4-D2.5 mg/L+NAA 0.3 mg/L。适宜的激素和非激素组合,能促进虎杖获得质地疏松、淡黄色、生长旺盛的愈伤组织。
Study on optimum factors of loose callus induction of Polygonum cuspidatum. By single factor and orthogonal experiments, the effects of different explants,exhormonal and hormonal combinations on callus induction, loosen and growth of Polygonum cuspidatum were investigated. Results showed that Leafstalk was much easier to induct loose callus induction, the optimum exhormonal factors of callus induction were pH 5.8, sucrose 5 g/L, casein hydrolysate 300 mg/L, inositol 10 mg/L, the optimum hormonal combinations of growth of Polygonum cuspidatum were 2,4-D 2. 5 mg/L ＋NAA 0.3 mg/L. The loosen, yellow, growth bloom callus induction of Polygonum cuspidatum were obtained under the proper exhormonal and hormonal.
