摘要
利用叶绿体基因trnS-trnG基因间隔区的通用引物,扩增并分析了不同地区的14个叶籽银杏和2个银杏的trnS-trnG基因间隔区序列,结果表明,14个叶籽银杏和2个银杏的trnS-trnG序列长度均为1005bp,A+T的含量为62.19%~62.39%。用DNASTAR软件对14个叶籽银杏和2个银杏的trnS-trnG基因间隔区序列进行同源性和差异性分析,建立了系统进化树。各样品之间变异小,14个叶籽银杏和2个银杏共有23个可变位点,1个信息位点,这些变异都以碱基的置换发生。除这些变异位点外的核苷酸序列完全相同,说明各样品序列之间具有高度同源性。YZ1变异位点较多,与其它样品的同源性较低。所测序列Blast检验后发现叶籽银杏trnS-trnG序列与银杏的相似度最高,苏铁类次之。
The cpDNA trnS-trnG intergenic spacer sequence of 14Ginkgo biloba var.epiphylla trees and 2 Ginkgo biloba trees which from different district were amplified and analyzed successfully.The results indicated that the size of every trnS-trnG sequences was 1005bp and the content of(A+T)% was 62.19%~62.39%.DNASTAR software was used to analyse homology and diversity between sequences and construct phylogenetic tree based on all the trnS-trnG sequences.There were 23 variable sizes and one information size in this 14 Ginkgo biloba var.epiphylla trees and 2 Ginkgo biloba trees,these variation all occurred in base substitution.Among the trnS-trnG sequences had little difference.YZ1 had more variable sizes and low homology with other samples.All the sequences through Blast found that it had the nearest relationship with Ginkgo biloba,Cycas the next one.
