摘要
本文应用原位杂交组化方法研究了降钙素基因相关肽(CGRP)的。α-和β-两种亚型的mRNA及编码P物质(SP)的前原速激肽AmRNA(PPTAmRNA大鼠三叉神经节神经元中的表达状况。结果发现:大约有35.7%,22.3%和17.5%的三叉神经节神经元分别有α-CGRP,β-CGRP和PPTA-mRNTA表达。中、小型神经元含有上述三种肽的mRNAs,而大神经元则主要表达。α-CGRPmRNA,从而提示它们在感觉信息的传递过程中可能起着不同的作用。相邻切片法研究的结果发现:这三种肽的mRNAs可共同表达于一些中、小神经元中,提示SP和CGRP在功能上的密切关系。
Calcitonin gene-related peptide (CGRP) and substance P (SP) are two of the most ubiquitousPeptides found in the cells of sensory ganglia, including the trigeminal ganglia (TG), indicating the importantroles for them in the transmission of sensory information. Recently, two kinds of CGRP, namely α- andβ-CGRP, and preprotachykinin A (PPTA) which coded for SP have been found in the brain by molecular clon-ing analysis. However, whether or not the two CGRPs and SP mRNAs are expressed in the same or differentcells in the TG is not yet clear. In the present study, in situ hybridization was used to differentiate these threekinds of mRNAs in neurons of the rat TG. In general, about 35.7 %, 22.3 % and 17.5 % of the TG cells were la-beled by α-CGRP, β-CGRP and PPTA probes, respectively. Small and medium-sized TG cells expressed allthese three types of mRNAs, but the large cells mainly contained the a-CGRP mRNA. The genes coding forthese Peptides coexisted in same small or mediumr-sized neurons. The present study showed that u-CGRP,P--CGRP and SP mRNAs were expressed in different neurons of the TG, indicating the different roles for themin the primary sensory system. The colocalization of CGRP and SP mRNAs in single neuron of the TG sug-gested a close functional relationship between CGRP and SP in the sensory transmission system.
