摘要
以西洋杜鹃的嫩叶片为外植体,研究了其离体培养和植株再生的过程。结果表明:叶片愈伤组织诱导最佳培养基为Read+ZT 5.0mg/L,可用回归模型y=19.21+7.32x_1来估测西洋杜鹃叶片愈伤组织诱导情况。叶片愈伤组织增殖培养基同初代培养基,继代30 d后体积可增为原来的3倍,重量增为原来的2.5~4.5倍。愈伤组织分化的最佳培养基配方为:Read+ZT 1.0 mg/L+NAA 0.01 mg/L,分化率为87.5%,分化出苗数在12~18株。生根培养基以Read+IBA 1.0 mg/L+NAA2.0 mg/L+ZT0.2 mg/L为好,生根率达90%以上。炼苗基质选腐殖土:黄沙:珍珠岩=5:1:1为宜,存活率达88.33%。
The technique system of induction of callus from leaf explants of Rhododendron hybridum and its regeneration were established. The results showed that Read + ZT 5.0 mg/L was the best medium to induce callus from leaf explants. And Linear regression modely = 19.21 + 7.32 x1 could estimate the growth condition of callus. Multiplication medium was the same as induction medium of callus. After 30 d inoculated, callus reached 3 times of its volume,2.5 -4.5 times of its weight. Read + ZT 1. 0 mg/L + NAA 0.10 mg/L was the best medium inducing callus to differentiate shoots and a high frequency could reach 87.5%. The regenerated shoots rooted readily on Read medium plus 1.0 mg/L IBA 2.0 mg/L NAA and ZT 0.2 mg/L. The rate of roofing could reach 90%. The regenerate plants were acclimatized the soil of muck, yellow sand and perlite (5: 1: 1) ,the survival rate of plantlets was 88.33%.
