摘要
以桃叶杜鹃无菌种子为试材,对影响其愈伤组织诱导和增殖的几个关键因子进行了研究。结果表明:无菌种子萌发过程中不需要添加植物激素,1/4 MS空白培养基即可获得理想的萌发率;在愈伤组织诱导中,MS培养基较其他培养基效果好,TDZ的诱导效果较其他细胞分裂素好,当TDZ 1.5 mg/L配以NAA 0.5 mg/L时,诱导率最高;在愈伤组织增殖中,TDZ 0.5 mg/L配以NAA 0.1 mg/L效果最好,增殖率为85%,增殖倍数为4.0;在培养基中添加水解乳蛋白能极大抑制愈伤组织褐化和促进愈伤组织的增殖。
Using seeds as experiment material,the several key factors on callus of Rhododendron annae induction and multiplication in tissue culture were studied.The results showed that 1/4 MS medium without hormone was more suitable than others for seed germination in aseptic culture.MS medium was the best medium for induction and multiplication of callus.The MS medium supplemented with TDZ 1.5 mg/L and NAA 0.5 mg/L had high rate on callus induction.The MS medium supplemented with TDZ 0.5 mg/L and NAA 0.1 mg/L was suitable for callus multiplication,and the multiplication rate could reach 85 percent,the multiplication ratio was 4.0.In order to avoid browning of callus,hydrolytic lactalbumin was supplemented in MS medium.It could improve callus multiplication.
